Neutron diffraction experiments have been performed on a magnetically ordered CeCu2Si2 single crystal exhibiting A-phase anomalies in specific heat and thermal expansion. Below T(N) approximately 0.8 K antiferromagnetic superstructure peaks have been detected. The propagation vector of the magnetic order appears to be determined by the topology of the Fermi surface of heavy quasiparticles as indicated by renormalized band-structure calculations. The observation of long-range incommensurate antiferromagnetic order as the nature of the A phase in CeCu2Si2 suggests that a spin-density-wave instability is the origin of the quantum critical point in CeCu2Si2.  相似文献

Background

As development proceeds the human embryo attains an ever more complex three dimensional (3D) structure. Analyzing the gene expression patterns that underlie these changes and interpreting their significance depends on identifying the anatomical structures to which they map and following these patterns in developing 3D structures over time. The difficulty of this task greatly increases as more gene expression patterns are added, particularly in organs with complex 3D structures such as the brain. Optical Projection Tomography (OPT) is a new technology which has been developed for rapidly generating digital 3D models of intact specimens. We have assessed the resolution of unstained neuronal structures within a Carnegie Stage (CS)17 OPT model and tested its use as a framework onto which anatomical structures can be defined and gene expression data mapped.

Results

Resolution of the OPT models was assessed by comparison of digital sections with physical sections stained, either with haematoxylin and eosin (H&E) or by immunocytochemistry for GAP43 or PAX6, to identify specific anatomical features. Despite the 3D models being of unstained tissue, peripheral nervous system structures from the trigeminal ganglion (~300 μm by ~150 μm) to the rootlets of cranial nerve XII (~20 μm in diameter) were clearly identifiable, as were structures in the developing neural tube such as the zona limitans intrathalamica (core is ~30 μm thick). Fourteen anatomical domains have been identified and visualised within the CS17 model. Two 3D gene expression domains, known to be defined by Pax6 expression in the mouse, were clearly visible when PAX6 data from 2D sections were mapped to the CS17 model. The feasibility of applying the OPT technology to all stages from CS12 to CS23, which encompasses the major period of organogenesis for the human developing central nervous system, was successfully demonstrated.

Conclusion

In the CS17 model considerable detail is visible within the developing nervous system at a minimum resolution of ~20 μm and 3D anatomical and gene expression domains can be defined and visualised successfully. The OPT models and accompanying technologies for manipulating them provide a powerful approach to visualising and analysing gene expression and morphology during early human brain development.  相似文献

A semi-Markovian point process which qualitatively models platooned arrivals is introduced. This process is used as the input to a single server queue in which the service times are independent and have a common distribution of phase type.It is shown that this queue has an embedded Markov chain of a particular block-partitioned type, whose invariant probability vector in the stable case is of matrix-geometric form. Detailed algorithms for the computation of the steady-state features of the queue are obtained and a representative numerical example is discussed.  相似文献

Various substrates bound to polyethylene glycol (PEG) through succinyl ester linkages were cleaved under catalytic transfer hydrogenation conditions. An experimental investigation has been carried out to measure the size of nano-aluminium agglomerates emerging from the combustion of nano-aluminized sandwiches and composite solid propellants. Nano-aluminium of median size of 50 nm produced in-house by the electrical wire explosion method is used in these samples. Propellants with different sizes of coarse and fine ammonium perchlorate are considered. Surface features of sandwiches and a propellant whose burning was interrupted by rapid depressurization are examined in a scanning electron microscope. The combustion products of the sandwiches and propellants are quenched close to the burning surface and collected in a quench collection set-up. The surface features of rapid-depressurization quenched sandwiches exhibit relatively large nano-aluminium clusters—of the order a few micrometres—particularly in the binder lamina. Quench-collected nano-aluminium exhibits significant agglomeration, but only a small fraction of the agglomerates are in the 1–3 μm range, except for both the coarse and fine AP particles used in the formulation being large, but even there they do not exceed ∼5 μm in size. This is expected to be benign for reduced smoke propellant applications from exhaust signature point of view, and to decrease the specific impulse losses without sacrificing the energetics of the propellant.  相似文献

We carried out specific-heat and ac-susceptibility experiments under hydrostatic pressure to investigate the interplay of spin-density-wave antiferromagnetism (A) and superconductivity (S) in single-crystalline AS-type CeCu(2)Si(2). We find evidence for a line of magnetic-field- and pressure-tuned quantum critical points in the normal state in the zero-temperature magnetic field-pressure plane. Our analysis suggests an extension of this line into the superconducting state and corroborates the close connection of the underlying mechanisms leading to the formation of the antiferromagnetic and the superconducting states in AS-type CeCu(2)Si(2).  相似文献

A highly sensitive and specific atmospheric pressure chemical ionization liquid chromatography-tandem mass spectrometry method was developed for serum pharmacokinetic studies of puerarin in rats. Chromatography was carried out on a reversed-phase Phenomenex Synergi 4 microm Fusion-RP80 column (150 x 2.0 mm i.d.) using a mobile phase consisting of acetonitrile-water (10:90, v/v) in 10 mm NH(4)OAc with a flow rate of 0.2 mL/min. Puerarin was analyzed in the multiple reaction monitoring mode with a precursor/product ion transition of m/z 415/267. The method was demonstrated to be specific and sensitive, and a linear response was observed over a range of 2-5000 ng/mL in rat serum. The validated method was successfully applied to the characterization of the pharmacokinetics of puerarin in rat serum after oral administration to spontaneously hypertensive rats. The blood concentration-time profile of puerarin showed a rapid initial increase, reaching a maximum and then declining within 1 h. Puerarin could not be detected after 24 h. The main pharmacokinetic parameters for puerarin after oral administration were as follows: C(max) (3.54 +/- 2.03 mg/L), T(max) (0.68 +/- 0.37 h), AUC(0-t) (7.29 +/- 3.79 mg h/L), AUC(0-infinity) (9.17 +/- 4.87 mg h/L), T(1/2) (1.7 +/- 0.6 h), CL/F (7.24 +/- 4.27 L/h/kg) and V/F (17.88 +/- 13.55 L/h/kg).  相似文献

Denitrification was compared between Paracoccus sp. and Diaphorobacter sp. in this study, both of which were isolated from activated sludge of a denitrifying reactor. Denitrification of both isolates showed contrasting patterns, where Diaphorobacter sp. showed accumulation of nitrite in the medium while Paracoccus sp. showed no accumulation. The nitrate reduction rate was 1.5 times more than the nitrite reduction in Diaphorobacter sp., as analyzed by the resting state denitrification kinetics. Increasing the nitrate concentration in the medium increased the nitrite accumulation in Diaphorobacter sp., but not in Paracoccus sp., indicating a branched electron transfer during denitrification. Diaphorobacter sp. was unable to denitrify efficiently at high nitrate concentrations from 1 M, but Paracoccus sp. could denitrify even up to 2 M nitrate. Paracoccus sp. was found to be an efficient denitrifier with insignificant amounts of nitrite accumulation, and it could also denitrify high amounts of nitrate up to 2 M. Efficient denitrification without accumulation of intermediates like nitrite is desirable in the removal of high nitrates from wastewaters. Paracoccus sp. is shown to suffice this demand and could be a potential organism to remove high nitrates effectively.  相似文献

A thin-layer chromatographic (TLC) procedure has been developed for the determination of astemizole in plasma as the free and as protein-bound substance. The detection and quantification were performed without using internal standards. In earlier described methods for the estimation of astemizole by high-performance liquid chromatography and radioimmunoassay, only free levels in plasma were quantified, at 3.3% of the total astemizole, with the remaining 96.7% bound to plasma protein and tissue. Our method employs proteolysis of plasma proteins by incubating plasma for 2 h in pepsin. After proteolysis the astemizole is extracted, and a known amount of the extract is spotted on precoated silica gel F 254 plates. Astemizole was quantified using a Shimadzu CS-930 dual-wavelength TLC scanner. The method provides a direct estimate of total astemizole present in the plasma.  相似文献

We present a novel simulation algorithm based on tempering a fraction of relaxation-limiting interactions to accelerate the process of obtaining uncorrelated equilibrium configurations of self-associating polymer solutions. This approach consists of tempering (turning off) the attractive interactions for a fraction of self-associating groups determined by a biasing field h. A number of independent configurations (replicas) with overlapping Hamiltonian distributions in the expanded (NVTh) ensemble with constant NVT but different biasing fields, forming a chain of Hamiltonians, were simulated in parallel with occasional attempts to exchange the replicas associated with adjacent fields. Each field had an associated distribution of tempered interactions, average fraction of tempered interactions, and structural decorrelation time. Tempering parameters (number of replicas, fields, and exchange frequencies) were chosen to obtain the highest efficiency in sampling equilibrium configurations of a self-association polymer solution based on short serial simulation runs and a statistical model. Depending on the strength of the relaxation-limiting interactions, system size, and thermodynamic conditions, the algorithm can be orders of magnitude more efficient than conventional canonical simulation and is superior to conventional temperature parallel tempering.  相似文献
